2 MATERIALS AND METHODS 2.1 Extract preparation Velvetbean Mucuna pruriens DC var. utilis Wight ; Burck PI365315 01 SD origin Mozambique ; was grown out-of-doors in pots containing 38 L of Krome gravelly loam soil at the Tropical Research and Education Center, University of Florida, Homestead, FL. Two months after planting, the main stem of each plant was cut at ground level, and the soil was washed gently from the roots. The plants were partitioned into main and fine roots, leaf blades, petioles and vines. At equivalent oral doses, cefadroxil has a longer serum half-life, slower urinary excretion rate, greater area under the serum level versus time curve than cephalexin or cephradine, and peak serum concentrations that are 75 to 80% those of cephalexin. The calculated, apparent in vivo volume of distribution of cefadroxil is greater than that of cephalexin. These properties infer greater persistence of cefadroxil in serum and urine and more prolonged in vivo bacterial exposure to cefadroxil than to cephalexin or cephradine. Neither cefadroxil nor cephalexin demonstrates drug accumulation on repeated administration. The serum levels achieved by cefadroxil are unaffected by food. The pharmacokinetic properties of cefadroxil are supportive of the development of clinical efficacy data which could indicate that cefadroxil could be administered at 12-h intervals. The studies reported here were undertaken to compare the pharmacokinetic properties in humans of the orally administered cephalosporin antibiotics cefadroxil, cephalexin, and cephradine structures presented in Fig. 1 ; and to determine the factors which control their disposition in vivo. Risk of VTE is 3- to 4-fold higher in women who take combined OCs containing less than 50 g EE per dose than in non-users.56 A comparison of the risks of VTE with different combined OCs found no evidence of any difference in risk between formulations containing less than 50 g EE.57 Interestingly this study determined that patient characteristics had more of an effect on risk, with smoking, body mass index over 35, asthma, and general ill health significantly increasing VTE risk. Risk of VTE also increases with aging.56 For example, combined OC users between the ages of 20 and 24 have an incidence of approximately 9 events per 10, 000 women per year, as compared to 18 events per 10, 000 women per year in combined OC users between the ages of 40 and 44.49 In 1995, concern was raised that third-generation progestins increased risk of VTE 2-fold more than older progestins. This was based on epidemiological studies. Data from additional studies have not shown this difference in risk.11 Similarly, although concern has been raised about an increased risk of VTE with combined OCs containing cyproterone acetate compared to other combined OCs in current use, a review of six epidemiological studies concluded that this is not the case.58 6.2.2 Myocardial infarction and stroke Cardiovascular effects of combined OCs are believed to be due to the estrogen component in the formulation, which may cause a dose-related stimulation of thrombosis.5 The reduction in dose of EE in combined OCs to below 50 g per tablet has eliminated excess risk of myocardial infarction MI ; , ischemic stroke, and hemorrhagic stroke for healthy, normotensive, non-smoking women.7, 44, 59 It is the presence of other risk factors for MI and stroke that affect overall risk to a much larger extent than combined OC use.

Section 442.6 of the Code of Federal Regulations 21 CFR 442.6 ; sets forth standards a monograph ; for the identity, strength, quality, and purity found necessary to adequately ensure the safety and efficacy of products containing cefadroxil monohydrate and test methods to determine compliance with such standards. You state that the monograph for bulk cefadroxil monohydrate 21 CFR 442.6 ; specifies that the product must be a * 'monohydrate and that its moisture content must be between 4.2 and 6.0 percent. You further state that the monographs for cefadroxil monohydrate capsules, tablets, and oral suspension 21 CFR 442.106a-c ; incorporate by reference the requirements of the monograph for the bulkdrug. You assert that, based on an analysis of a sample of the bulk cefadroxil product manufactured by Gema and provided by Zenith to Bristol-Myers Squibb in a court proceeding, the product is not a monohydrate'. Therefore, it does not comply with the monograph at 21 CFR 442.6 and the applications for approval sought by Gema and Zenith must be denied and lincocin.
